From: Regulation of cell proliferation and cell density by the inorganic phosphate transporter PiT1

Figure 6

Over-expression of hPiT1 increases proliferation and cell density of NIH3T3 cultures. A) Cell counts. Control (NIH3T3-LXSN (LXSN)) (squares) and NIH3T3-LPiT1SN (LPiT1SN) (crosses) cells were seeded at 20,000 cells/cm2 in 4-well plates and counted at the indicated days. The results are shown as mean cell number per cm2 of 4 wells ± SD. *indicates statistically significantly different from NIH3T3-LXSN cells at the same day, p < 0.05. B and C) Cell cycle analysis. NIH3T3-LPiT1SN (LPiT1SN) and NIH3T3-LXSN (LXSN) were seeded at 20,000 cells/cm2 in T25-flasks, and cultures were stained with propidium iodide 8, 12, and 24 hours later, and analyzed by flow cytometry. B) Representative flow cytometry histograms of NIH3T3-LXSN (LXSN) (left) and NIH3T3-LPiT1SN (LPiT1SN) (right) cells at 24 hours after seeding; the thick black lines represent the Watson-Pragmatic model. C) The percentages of cells in the respective phases of the cell cycle 8, 12, and 24 hours after seeding as indicated obtained using the Watson Pragmatic model. Each column represents the mean of triplicate set ups ± SD. *indicates statistically significantly different from NIH3T3-LXSN (control) cells in the same phases of the cell cycle, p < 0.05.
