Phosphorylation of TBX3 at SP190 reduces its ability to promote cell proliferation in a p21-dependent manner. Upper panel SW1353 shCtrl and shTBX3 cells were co-transfected with pCMV empty, WT TBX3 (pCMV-TBX3), TBX3 S190A (pCMV-S190A) or TBX3 S190E (pCMV-S190E) expression constructs. Twenty four hours following transfection cells were replated at 4000 cells/well in 96 well plates and allowed to adhere for 24 h. Cells were subjected to MTT and metabolization measured at OD 595 nm. The values indicate the mean of three independent experiments ± SEM (**p < 0.01 and ***p < 0.001). Lower panel western blotting and qRT-PCR showing a correlation between the expression of WT TBX3, TBX3 S190A and TBX3 S190E and p21 protein and mRNA levels in SW1353 shTBX3 cells.
