Dynamic regulation of CRLs activity by neddylation and deneddylation. CAND1 binding to unmodified cullin blocks the interaction of cullin with the substrate receptor-adaptor module. Cullin neddylation promotes the dissociation of cullin from CAND1 and restores the CRLs in an active conformation, leading to substrate ubiquitination. After the detachment of ubiquitinated substrate from CRLs, NEDD8 is removed by CSN from cullin for recycling. At last, CAND1 binds to cullin and inactivates CRLs
